Virusi - Zgradba, Pomen in Vpliv na Človeka in Ekosistem

Zgradba virusov – kaj so ti skrivnostni delci?
Virusi so mikro delci, manjši od bakterij, ki niso prave celice. Zato jih ne štejemo med živa bitja v klasičnem smislu – so kot nekakšni "biološki roboti" na meji med živim in neživim. Brez gostiteljske celice so popolnoma neaktivni, kot kristali.
Vsak virus ima osnovno zgradbo iz dveh delov. Dedni material je lahko DNK ali RNK (nikoli oba hkrati!), ki nosi navodila za izdelavo novih virusov. Okoli njega je kapsida – beljakovinski plašč, ki deluje kot zaščitni ovoj.
Nekateri virusi imajo še dodatno virusno ovojnico z glikoproteini na površini. Ti delujejo kot ključi, s katerimi virus "odklene" gostiteljsko celico. Glede na obliko kapside ločimo paličaste viruse (kot tobačni mozaik), poliedrične (npr. adenovirusi) in kompleksne (bakteriofagi z "glavo" in "repkom").
Pomembno za test: Virus ima samo DNK ali samo RNK, nikoli obeh hkrati!
Litični cikel – hitro in uničuječe
Litični cikel je kot virusni blitzkrieg – hiter in smrtonosen za gostiteljsko celico. Virus se najprej pritrdi na specifične receptorje celice, nato vbrizga svoj dedni material vanjo.
Ko je virus znotraj, prevzame popoln nadzor nad celico. Celica pozabi na svoje naloge in začne kot prava tovarna proizvajati virusne komponente – nove nukleinske kisline in beljakovine za kapside. Ti deli se spontano sestavijo v na stotine novih virusov.
Končni rezultat je spektakularen: celica dobesedno poči (liza) in sprosti na stotine novih virusov, ki takoj okužijo sosednje celice. Gostiteljska celica pri tem seveda umre. Tipični primeri so virus gripe in prehlada, ki povzročijo hitre simptome.
Ključno: Pri litičnem ciklu celica hitro umre in sprosti kopico novih virusov!
Lizogeni cikel – skriti sovražnik
Lizogeni cikel je popolnoma drugačen – virus deluje kot špion, ki se skrije in čaka na pravi trenutek. Virusna DNK se vgradi v kromosom gostiteljske celice in postane provirus (pri bakteriofagih se imenuje profag).
Ta vgrajeni virus je popolnoma "tiho" in se obnaša kot del celičnega genoma. Vsakič, ko se celica normalno deli, se skupaj z njo prekopira tudi virusni dedni material. Tako se virus širi, ne da bi povzročil kakršno koli škodo.
Toda ta mirnost ni trajna! Zaradi različnih sprožilcev (UV sevanje, stres, kemikalije) se lahko provirus aktivira in nenadoma preide v litični cikel. Takrat začne množična proizvodnja virusov in celica propade.
Klasični primeri so herpes virusi, HIV in virus noric . Pri herpesih se virus lahko skriva leta v živčnih celicah, nato pa se aktivira ob stresu ali oslabljeni imunosti.
Pozor na test: Lizogeni cikel = virus se skrije v genom in čaka!
Primeri virusov v praksi
Virus gripe je RNK virus z ovojnico, ki napade celice dihal z litičnim ciklom. Njegova supermočnost je izjemno hitro mutiranje – zato potrebujemo novo cepivo vsako leto. Simptomi se pojavijo hitro: vročina, kašelj, bolečine v mišicah.
Bakteriofag T4 je pravi "robot" med virusi s kompleksno zgradbo – ima "glavo" polno DNK in "repek" za injiciranje. Napade bakterijo E. coli in je odličen model za študij obeh ciklov razmnoževanja.
HIV virus je še posebej zahrbtен retrovirus. Ima encim reverzno transkriptazo, ki prepiše virusno RNK v DNK. Ta se vgradi v genom imunskih celic in tam "spi" leta, nato pa se aktivira in uniči imunski sistem, kar povzroči AIDS.
Ključna razlika: Retrovirusi lahko RNK prepišejo v DNK – to je obrnjen proces!
Virusi in človek – sovražniki ali zavezniki?
Antibiotiki proti virusom ne delujejo! To je ena najpomembnejših stvari, ki jih moraš vedeti. Antibiotiki uničujejo bakterijske celične stene, virusi pa celic nimajo. Proti virusom uporabljamo antivirotike ali se zaščitimo s cepljenjem.
Cepljenje je najučinkovitejša obramba – v telo vnesemo oslabljen virus ali le njegov del, da imunski sistem razvije protitelesa in "pomni" napadalca. Ko pride pravi virus, je imunski sistem pripravljen.
So virusi živi? To je ena najbolj zanimivih razprav v biologiji. Imajo dedni material in se razmnožujejo (z evolucijo), a nimajo celične zgradbe in lastne presnove. Zaključek: so na fascinantni meji med živim in neživim svetom.
Virusi niso samo škodljivci! V genskem inženiringu jih uporabljamo kot "dostavna vozila" za vnašanje genov. Fagna terapija z bakteriofagi postaja alternativa antibiotikom proti odpornim bakterijam.
