Kaj so mutacije in osnovni pojmi
Mutacije so trajne spremembe v dednem materialu - torej v tvoji DNA. Dogajajo se povsem naključno in v katerikoli celici, kar pomeni, da je vsak od nas genetsko edinstven.
Te spremembe so kot "tipkarske napake" v veliki knjigi življenja. Nekatere so nevtralne, druge škodljive, redke pa celo koristne. Brez njih ne bi bilo evolucije!
Mutageni so dejavniki, ki povečajo verjetnost mutacij - UV sevanje, cigaretni dim ali nekateri virusi. Kariotip pa je kot "katalog" vseh tvojih kromosomov, urejen po velikosti in obliki.
💡 Zanimivost: Tvoja DNA se kopira milijonkrat v življenju, zato so mutacije v resnici neizbežne - na srečo imamo odličen "korektorski" sistem!
Genske mutacije - spremembe na nivoju genov
Pri genskih mutacijah se spremeni zaporedje baz v DNA. Obstajajo trije glavni tipi, ki imajo različne posledice.
Substitucija pomeni zamenjavo enega nukleotida. Lahko je tiha (brez posledic), napačnega smisla (druga aminokislina) ali nesmiselna (prezgodnji stop signal). Slednja je najhujša, ker nastane okrnjen protein.
Insercija (vstavitev) in delecija (izguba) nukleotidov sta še posebej problematični. Če število ni večkratnik 3, pride do premika bralnega okvira - od tega mesta naprej se vsi kodoni berejo napačno.
💡 Pomni: Frameshift mutacije so skoraj vedno katastrofalne - kot da bi v stavku "KAT JE DOB ER" vstavil črko in dobil "KXAT JE DOB ER"!
Kromosomske mutacije - večje spremembe
Kromosomske mutacije prizadenejo cele dele kromosomov ali kar cele kromosome. Te spremembe so vidne pod mikroskopom pri analizi kariotipa.
Strukturne mutacije vključujejo delecijo (izguba dela), duplikacijo (podvojitev dela), inverzijo (obrat za 180°) in translokacijo (prenos na drug kromosom). Predstavljaj si kot prestavljanje delov puzzle.
Numerične mutacije pomenijo napačno število kromosomov. Trisomija je prisotnost treh kopij namesto dveh (npr. Downov sindrom), monosomija pa pomanjkanje enega kromosoma. Poliploidija pomeni dodatne cele komplete kromosomov.
💡 Zanimivost: Poliploidija je pri živalih običajno smrtna, pri rastlinah pa pogosta - pšenica in jagode so poliploidne!
Konkretni primeri mutacij
Anemija srpastih celic je odličen primer genske mutacije. Zamenjava enega samega nukleotida povzroči, da se glutaminska kislina zamenja z valinom v hemoglobinu. Rezultat? Rdeče krvničke dobijo obliko srpa in slabše prenašajo kisik.
Downov sindrom je primer kromosomske mutacije - trisomija 21. Oseba ima tri kopije 21. kromosoma namesto dveh, skupno torej 47 kromosomov. To povzroči značilne telesne lastnosti in motnje v razvoju.
Vzrok je običajno nerazdvajanje kromosomov med mejozo, ko se spolne celice ne razdelijo pravilno. Tveganje se povečuje z materino starostjo.
💡 Pomembno za teste: Mutacije v telesnih celicah (npr. rak) niso dedne, mutacije v spolnih celicah pa se prenašajo na potomce!
Vzroki mutacij in njihov pomen
Spontane mutacije nastajajo kot napake pri podvojevanju DNA. So redke, ker ima DNA-polimeraza odličen "korektorski" sistem - kot preverjanje črkovanja pri pisanju.
Inducirane mutacije povzročajo mutageni: UV sevanje (kožni rak), rentgenski žarki, snovi v cigaretnem dimu, azbest ali nekateri virusi kot HPV. Pomembno je poznati glavne vire!
Mutacije so motor evolucije - brez njih bi bili vsi organizmi identični. Večina je nevtralnih ali škodljivih, redke koristne pa dajejo prednosti v določenem okolju. Odpornost bakterij na antibiotike je odličen primer koristne mutacije.

Hiter povzetek za ponavljanje
